Tao et al. show that aging mouse colon-derived organoids spontaneously silence genes, which activates the Wnt pathway and makes them more sensitive than young ones to BrafV600E-induced transformation. Simultaneous deletion of a panel of the silenced genes sensitizes young organoids to transformation by BrafV600E.
