By combining biallelic C/EBPα and GATA-2 ZnF1 mutations, Di Genua et al. generate a mouse model of bilineage acute erythroid leukemia and identify a neutrophil-monocyte progenitor (NMP) that undergoes transcriptional and epigenetic reprogramming to express erythroid genes as the major leukemia-initiating cell.
